To burn a file of this size it is necessary to use the UDF format:
http://es.wikipedia.org/wiki/Universal_Disk_Format
In Debian, we install the udftools package to be able to give this format to the .iso file that we will burn.
For example:
apt-get install udftools
dd if=/dev/zero of=file.iso bs=1024 count=4589800
mkudffs file.iso
mkdir /mnt/loop
mount -o loop -t udf file.iso /mnt/loop
cp big_file /mnt/loop
umount /mnt/loop
growisofs -speed 4 -Z /dev/dvd=file.iso
